| The hsp70 promotor (from 87C1), extending from the XbaI site at -245 to the XmnI site at +207, was inserted upstream from the actin 5C 3' trailer and poly(A) addition sites. This is an 800 bp fragment excised from pCaSpeR-act (see Thummel et al., Gene, 74: 445-456). The Act5C 3' region will allow the encoded mRNAs to be stable under non-heat shock conditions. A polylinker containing 8 unique sites (underlined) can be used to insert an open reading frame for expression. | |
